Our interpretation.

This is a burst of comic coaching: if anyone asks whether you're a god, the speaker insists the answer is simply yes. It plays on how absurdly bold that would be, and on the urge to grab a chance when it appears.

In the temple on top of Dana's apartment building, Gozer appears to the Ghostbusters in the form of a woman, and Ray formally orders it, on behalf of the City, County and State of New York, to return to its place of origin. Gozer asks him if he is a god; Ray glances at Venkman and answers no, and Gozer replies by blasting them all with lightning. This is Winston's advice, delivered while they are all still sprawled on the roof.
